DNA methylation is one of epigenetics, and aberrant DNA methylation is related to various diseases such as cancer, neurological diseases, and allergic diseases. It is necessary to understand the methylation status of DNA in order to elucidate the fundamental cause of these diseases and to make an accurate diagnosis. Currently, the bisulfite sequencing method (BS-seq) is generally widely used as a DNA methylation analysis method. However, BS-seq has a problem that it cannot distinguish between hydroxymethylated cytosine (hmC) and methylated cytosine (mC). Therefore, our laboratory is trying to develop a new methylation analysis method that combines DNA (cytosine-5) methyltransferase 1 (Dnmt1) and Polymerase. In this study, recombinant Dnmt1 with low de novo activity and low activity against hmC in vitro was prepared and applied to the methylation analysis of the tyrosinase promoter of B16 melanoma. As a result, the difference in the methylation site was observed between the analysis result of only BS-seq and the analysis result of the novel methylation analysis method. From this result, it is considered that by combining BS-seq with recombinant Dnmt1, only methylation could be detected excluding hydroxymethylation. |
